DCA schedule in C6 rat glioma model

Interesting Ukrainian study showing schedule-dependent effects of DCA.  Daily DCA treatment for 6 days starting at day 2 decreased average life span by 15%, daily treatment for 6 days starting at day 7 had no effect on survival.  Longer treatment (13 days) starting at day 2 increased average life span by 25% or more.  The DCA treatment was more effective under hypoxic conditions.

  1. Stephen, does that mean you probably shouldn't use DCA and Chloroquine together?

    1. My guess is that chloroquine and DCA are a good combination, as tumor cells in conditions of hypoxia are more vulnerable to the effects of both of these drugs.

  2. Do you think that unless you can tolerate DCA at higher doses for at least 2 weeks at a time there is probably little chance of it doing any good? My wife is a slow metabolizer and shows side effects at day 2 even after a long break at a low dose.

    1. What is the low dose she is taking?

Because of differences in metabolism some people will achieve the same plasma levels and the same benefit that others would only get with higher doses.

Did you start at a high dose and later reduce to a low dose after she developed symptoms? What was the starting dose?

    2. The latest schedule that she was on for 3 weeks was as follows:
      Day 1 400mg morning/300mg night
      Day 2 300mg morning/300mg night
      Day 3/4 200mg morning/200mg night
      Day 5/6/7 0mg
This schedule was started after a 2 week break and symptoms would start by day 2, hand tremors, disorientation, loss of balance. She is 60kg.
The first schedule we tried was 300mg once daily for 7 days followed by a week off.

  3. I can't find any information how to take DCA in relation to food? Before meals, during meals, after meals? Has anyone found this information?

    1. I wasn't able to find a pharmacokinetic study comparing DCA bioavailability with versus without food. But bioavailability is 100% after an overnight fast.

"an oral DCA dose is rapidly and
virtually completely absorbed, with peak plasma concentrations usually achieved within 15-30 min of administration to individuals fasted overnight"

  4. My mom now takes a DCA purity of 99%, but I doubt the quality of this product, since I bought it in a dubious place.

I would like to order a new DCA here:
    https://www.sigmaaldrich.com/catalog/product/aldrich/347795?lang=en&region=US
    assay 98%
    impurities <2% ethyl alcohol

What do you say? Is this what we need?
