Carboplatine + Avastin

Hi all,

Anyone had any sustained success with Avastin and carboplatine combination? Stephen, would appreciate your views please. We are struggling with decisions and my husband just had another recurrence less than 2 months after the last recurrence. Would carboplatine add any benefit? Did it show any efficacy? Many thanks.

  1. Unlike the BELOB trial (Avastin + CCNU [lomustine]), which showed superior results with combination treatment, a randomized phase 2 trial of Avastin + carboplatin (the CABARET trial) published in 2015 showed no significant difference in progression-free or overall survival when carboplatin was added to Avastin. Progression-free survival was virtually identical in both arms, and the Avastin alone arm had a slight but non-significant advantage in terms of overall survival.

"In this large, multicenter randomized phase 2 study,no obvious clinically significant benefit for the combination of bevacizumab and carboplatin was detected, and CABARET provides no support for further study of this combination."

  3. Thanks Stepen. He got Avastin again last Tuesday even though we knew about the recurrence then but we continued with treatment as is till we figure out an alternate plan., wouldCPT11 be a better choice? He failed temodar, CCNU, Metronomic Temodar so not sure what viable alternatives we are left with at this point?
