Polio/Rhino Virus

I know this isn't cocktail related, but CBS is doing a follow-up this Sunday. There is no question, a single therapy that eradicated recurrent GBM is stunning. I'm just wondering how we should view this? I don't believe there is such a thing as false hope. But, is this more media hype or a breakthrough? http://www.cbsnews.com/news/promising-duke-university-polio-brain-cancer-trial-given-breakthrough-status-60-minutes/

  1. Danny, I loved seeing this good news but (at least when we talked to Duke) it is not a solution for everyone. The virus is placed surgically and causes edema in most cases. So if you are inoperable or have had problem with edema, it might not be for you. On the other hand, those who can do it have nothing but blessings from me! Are you considering? We called Duke when we were looking at this and they got back to us quickly. Very helpful.

  2. This is definitely one of the most promising approaches, it can lead to long and durable responses in patients. It's not just Polio, both DNX-2401 and Toca511 look very good. Location is a big problem. Since these cause swelling in the brain, the tumor location is important for determining who can be included inn the trials. If it is located near the interior of the brain, then treatment is difficult because it could cause swelling near critical structures. Since the first trials, they have figured out proper dosage, so if a patient has a tumor in the optimal location it can work really well.

For people that do not qualify for this approach, immune checkpoint inhibitors might be good options but only in some patients.

    1. The idea behind Polio Virus vaccine is the winner. It works and I believe it has been and will continue to be improved upon. Duke also released very good survival statistics from early trials of their Dendretic Cell PP65 vaccine. DC PP65 has no known side effects. There is a current study testing PP65 along with Opdivo (Nivolumab). Will be interesting to see if adding a range of treatments (weapons) is effective. We are headed in the right direction.
