John cocktail

Full resection on Mar 27th 2015. Tumor is unmethylated TP53, no EGFR, no IDH1. Molecular report suggested mTOR inhibitors like Afinitor or Torisel. PDL1 is moderate. It says Giant Cell Glioblastoma.
During the radiation my brother took some CBD/THC but possible it was bad quality and might not have had any effect. The cocktail was started way after the radiation was over and some ingredients have been taken for about 2 months some have just been added.
I don't know what dosages is he taking since his wife is administering the drugs. First CT scan after radiation showed increased uptake of contrast. They just got PET SCAN results today and they don’t want to open it for another week (how irrational). He took local hyperthermia after the radiation.

  3. I might be wrong with this but after a few standard cycles of tmz we have just switched to some odd schedule. We wanted to make it metronomic but te pills were to hard to split so my brother is now taking one pill a day and all the pills are split over 28 days so 1 every 3 days or so.They don't prescribe metronomic tmz in poland. Doing so we hope that we will have at list a fraction of antiangiogenic activity, and also we hope that it will create more synergy with other drugs because for example if I am doing 100 mg of tmz a day for 5 days with a dose of 1000 of metformin per day that would give me 100x5000 killing power
    now if I spred it over the month I will do the same amount of tmz over the course of the month but with twice as much of metformin being in the blood at the same time with tmz and 2 times of other drugs. So my theory is that there will be more killing power and more synergy.

  4. I am cutting my 140mg pills into thirds to do my metronomic dosing. It's pretty easy. I got some more gel caps from the grocery store. And then you need a mirror, a razor blade and an 11 year old boy with a steady hand (make sure he washes his hands first). The tmz doesn't stay in your system for very long - hours not days. So taking it daily really might make a difference.

    1. If Temodar comes in capsules, how do you cut the capsule into thirds without having the TMZ spill out?

  5. If you are given TMZ at 200mg/m2 for 5 days, that is 1000 mg/m2 per cycle. If you divided that amount of TMZ evenly out over the 28 day cycle, that would equal about 36 mg/m2 per day. This is somewhat less than the standard metronomic dose of 50 mg/m2, but similar to what Rich took back in 2007 (40 mg/m2).

  7. Sorry Stephen. Didn't see your post until just now.
    If the TMZ is a fine powder in a capsule, twist the capsule open and dump the contents onto the mirror. Be careful, it can really make a mess. Save the capsule as you will use it. Take the razor blade and make a nice neat line and cut it into thirds.
    Cut one the lines long and skinny. If you can, elevate the mirror on erasers so that you can get half of one of the capsules under the mirror's edge (the eleven year old touch). Using the razor blade, carefully push the line into one of the capsule halves. Then twist the capsule halves back together. Repeat with your store bought capsules. If at any time you get frustrated or sneeze and the powder goes all over the place, yell at your boy. This is more effective if he is present, so don't yell at him too loud and he will go away and he won't be around for the next yelling. If you loose too much powder, let me know and I'll send you some more stuff.
    I noticed that the manufacture takes care with the packaging to protect the TMZ from radiation and moisture. I would be careful to do only a few capsules at a time and keep them out of the light).
