We are using 3 kinds of PPIs: Omeprazole, Lansoprazole and Esomeprazole to be had on a weekly basis as: 3 days high dose, 3 days low/standard dosem one day pause (=7 days); after a month we change to the next PPI. This is based on an informal protocol suggested by professor Stefano Fais. He has published on the order of 100 papers on PPIs and cancer, including the recent one on Lansoprazole. we had this as a recommendation from Anders on the virtualtrials mail group.. I use the high dose as: 2.5mg/kg/day.
Hi Sarah, I'm interested in following the PPI protocol as well. Are all 3 PPIs used at the same high does 2.5mg/kg/day. Also what is the low dose that you use. Thanks
Hi linda..sorry for late reply, yes for all PPIs. we calculate the high dose first..as i told u 2.5 mg/kg..say it is 5 pills..so the medium dose will be 2-3..and the low dose is usually 1 pill (the std dose).. I figured out this on my own..as i didnt recieve more details from Anders..

I know there was some talk if the omeprazole crosses the BBB. I have found some articles where It says that it does and that there is a rapid exchange etc.
I have passed the same dosage suggestions people described above to my sister in law who is administering the drugs to my brother but I am not sure if she is following it. I know that my brother has been on omeprazole for a few months but I don't know the exact dosage. In the past I contacted Anders Ferry and he said that omeprazole itself is like candy and there has not been any adverse events on people getting large doses of it for extended periods of time but in the context of such complex cocktails here you never know. My brother is about to switch to lansoprazole now, maybe did already.
