Chloroquine dose for EGFR Amplified/EGFRvIII positive GBM

Hello all, I just also reviewed this 1/17/2017 article regarding GBM and Chloroquine (https://www.sciencedaily.com/releases/2017/01/170117084050.htm) and there seems to be some efficacy with Chloroquine use in BRAF mutation positive GBM.  In our case the BRAF mutation was not detected and I'm wondering if CQ would still have a place in our cocktail in the absence of this mutation.  

However, as was pointed out on this blog, retrospective studies have indicated Chloroquine as being particularly effective in EGFRvIII positive GBM, which is the case with our GBM. I also noted clinical trials going on using CQ in GBM, but could not find any prelim data.  We just finished cycle 2 of TMZ and want to add Chloroquine to our cocktail right away.

Does anyone have an idea regarding the dose (mg) and how it should be taken? (one a day? twice a day? only during the 5/28?  Regards,

  1. Bahman, I've edited your post to insert an image. This was taken from an unpublished study which provided the rationale for a chloroquine trial currently underway for GBM.

    This retrospective study included patients from the published Mexico clinical trials (chloroquine in addition to radiation and BCNU chemotherapy), in which newly diagnosed GBM patients were give 150 mg chloroquine (equivalent to 250 mg chloroquine phosphate) daily. The schedule in this trial was as follows:

    "Chloroquine was administered at 150 mg/d, starting
    on day 5 after surgery, and was continued for 12 months"

    As chloroquine phosphate comes in doses of 250, this was likely given on a once daily schedule (250 mg chloroquine phosphate once daily = 150 chloroquine once daily).

  2. Our friend Rich (who was basically moderating the precursor of this blog at the "cocktails" thread on cancer compass), was taking the same dose of chloroquine phosphate (250 mg once daily) for nearly 2 years, in addition to numerous other drugs and supplements.

    1. Today we started CQ phos 500 mg half-tab daily which yields about 300 mg.

  5. Do you need a prescription for this? If not, how does one get it?

    1. Chloroquine is a prescription drug for malaria, but is in clinical trials for GBM and you may be able to find a doctor willing to prescribe it off-label.
